有关异染色质,最近发现两种不同的观点:
1、异染色质特指端粒、着丝粒、rDNA区域,占比大约是200M/3G(hg),也就是不到10%,异染色质并不等同于染色质非活跃区(https://bionumbers.hms.harvard.edu/files/DNA%20content%20of%20human%20chromosomes.pdf);
2、异染色质就是Compartment B,或者说Compartment B是异染色质,例如(https://doi.org/10.1093/bioinformatics/btx802)在Introduction中提到了“the B compartment corresponds to the heterochromatin regions associated with the nuclear lamina and nucleolar”;(https://www.nature.com/articles/s41467-021-22666-3)中Fig1下面的段落中提到“heterochromatin mark H3K9me3 in B.2.1 and B.2.2”(Kimura, 2013中曾经说H3K9me3 is broadly distributed on inactive regions);(doi: 10.3389/fmolb.2023.1168562)中也提到“H3K9me3—Histone 3 Lysine 9 tri-methylation is a chromatin modification typically associated with heterochromatin”,还有很多文献这里就不再赘述。我曾经计算过多个细胞系中Compartment B的占比都不低于20%,所以和观点1中说的比例也对不上
综上,我有点迷茫,异染色质到底是不是等同于Compartment B?还是说Compartment B仅是基因转录非活跃区(也就是说不一定是大尺度大片段)?对于第一个观点,是否还有其他证据?对于Compartment B究竟什么样子的描述才是准确的?求大佬指点!感激不尽!
